Nothing in the game can be missed. After finishing the game, you can load the last checkpoint and collect anything missed.

The first one is the December 19, 2020 build. The second one is the free 18+ Adult Only Content, which is required for one achievement.

Before starting, disable Steam Cloud and delete any existing saves (if any). The saves can be found in:

%appdata%\RenPy\Highway Blossoms Remastered-15500000
...\steamapps\common\Highway Blossoms\game\saves

You can re-enable Steam Cloud after confirming that the achievements are working.

Achievements fixed. Old version no longer required.

Settings → accessibility → More Difficulty Options: ON, Window Movement: OFF
Settings → wrench icon → P1 and P2 Difficulty: Unmissable

On Unmissable difficulty you can simple autoclick to win all tracks. However, 'Nimble' achievement (60 points in 2-B1) must be obtained legitimately as the related stage cannot be cheesed.

Additional Notes:

Fruits scattered throughout the game are optional and serve as a means to gain extra lives. It's best to avoid spending time or risking death to collect them.
Take the time to find all feathers/eggs and the dojo in one run in each level. If you die, you'll be reset to the last checkpoint, which can help for going back and retrieving any missed collectibles.
Each level has both a normal mode and a speed run mode. It's more efficient to aim for the gold medal in the speed run mode immediately after completing the level in normal mode. Since the layout and challenges will be fresh in your memory, you'll have an easier time achieving a faster completion.
You don't have to get the golden trophy version for the "Cataclysm mode" (no death). Just finish the level and take the time you need.

Buy all parts for the Camaro
Buy the Seagull

Skill based:
Deliver 5,000 points at once (Save cargo until you have 5000, then drive into green zone)
Get 10,000 points in a run
Collect 10 special packages
(These glow and stand out, but most likely require damage to be taken to be collected)

Dickland: Horror Quest (2436330)

Use A, D keyboard buttons to move left and right, W to jump, E to enter to the doors, ladders etc. F to interact and take items to your inventory, use arrows to choose an item in your inventory.

Go right, click E to your friend.
Go right, open the door and take the tool from there, then go right more and click E to your friend
Go right, use a tool to the tree, then talk with your friend, then go right and click E to complete level

Go right, take a part of the bridge, jump on the invisible platforms to right then left and then right, take one more part, interact with door and take one more part
Then go right until you reach wall, then fall left and go right to the river, wait until you can catch a part of the bridge, catch it and build it, go right press E

Fall right, jump into the left side wall, reach the end of the wall and jump spamming F to take a tool, get back right and use tool
Jump through the stumps and take a bucket ("F"). Use it on puddle, go further, when you will be between two paddles - use bucket to pour out the water to the left paddle and take water from the right paddle, repeat until reach end.

Take a log, go left, interact with door with this log (take a key), now you can open three containers using this key, take from there a one more key, open a last container, take a stick for the lever, insert it to the lever connector, use it and go left to open the door with second key
Go further avoiding enemies using the shelves.

Go up two times on the ladder, take a lever stick from the left wall, get back to the first floor, go left, insert a stick, open the gate, go left to the most left shelf, take a key from there
Take one more key from the container in the second floor avoiding enemy, go to the third floor, open container and take a tool, go to the first floor and use it on the stone between two shelves
Drop a rock from the third floor to the enemy, if you failed, you can take one more rock from the first floor, now way on the second floor is free, go left in the second floor and use a ladder
Interact with the curtain, take one more key and use it on the exit door

Enter the shelf, here you can find two more logs jumping and spamming F, reach the right shelf and take one more part from the room outside the right shelf
Get back to the first floor and build the ladder down on right from the monster, come down, take second key behind the door and third key behind the curtain, get back on the first floor, take a key from container
Get back to the room outside the right shelf, use a key, go further and use a key on the end of the level

Playthrough 1: Complete Game (~10 hours)

This can be done on any difficulty. I played on normal. Complete the game, including finishing all 16 sidequests, finding all 25 secret rooms, and recruiting the two optional party members (Beriberu and Magasz).

Some sidequests are time-limited, so it's important to start each quest immediately and complete them as soon as possible. They are listed in chronological order in the guide.

I don't think any secret rooms can be missed, but it's best to find them as you travel through each area. They're also listed in chronological order in the guide.

Playthrough 1.5: 20 Hours Played (~10 hours)

Continue from your latest save and stand in a safe place. Allow the in-game playtime to reach 20 hours. You can check the current playtime when you save. Note that the timer stops when the game window isn't active.

Playthrough 2: 6 Hour Playthrough (~3 hours)

It's 6 hours according to the in-game timer, so it's not hard at all. I played on easy. Only focus on the story and skip everything else.

Enslan joined my party at the 1-hour mark.
I grinded for an hour in the Outer Wasteland until level 35.
My last save before the final boss was at 2 hours 47 minutes.

- Run the script (press F1) on the main menu for approx. a minute.
- Delete saves in "%localappdata%\AchievementDisplayCabinet" if any missed unlocks.
- It's possible the sleep can be reduced, but with too low sleep I was missing several unlocks.

Cards and Castles 2

Notes:

Some grind to be able to craft needed cards is probably required, unless you are lucky in getting the below mentioned legendary cards. Or if you have enough currency from duplicates (especially "shiny" cards).
Go to "Shop", click on "gift code" and put in "steam" (might expire at some point)
Achievements pop at the end of a game.
The spectate achievement does not work when someone surrenders, so the game has to be played out.

Tips:

About "Big Swing - Destroy 10 or more enemy units in one turn in a multiplayer game.":
This only seems to work with the cleave ability (f.e. "Swordsman" card).
About "Monster Mash - Raise 10 or more zombies in a single play in a multiplayer game.":
The only way to do this seems to be with "Cemetery" x2 in both decks & "Necro-Commander Zethik" for the person who needs the achievement.
About "Massive Overkill - Deal 50 or more damage in a single attack in a multiplayer game.":
The damage needs to be done against ONE target only.
Example 1: use "Halls of Frost".
Example 2: Have a minimum of 11 skeletons on the board and then play "Bone Dragon Mordanoth". EDIT: The card seems to have been changed, and now it deals 3 damage times any skeleton on the board or on hand.
