Video of how to play Spain.
This will tell you what to expect out of the special units, what to expect out of the playstyle, and what victory conditions to expect to dominate with.

Victory Conditions
Hello, if you really didn't want to watch the video I'll give a quick rundown of what you probably should be thinking if you are randomly paired up with the Spanish or are just new to Civilization and think they would be fun to play!

Victory Conditions

  • Domination
Isabella is very cruel as an AI player who will usually always have fought you at least once in a playthrough. Should you do the same if you are playing as Spain? The answer to that is yes. Her special units are very good to go and attack someone with. The fact that you can use Conquistadors as Settlers so as long as it's in a foreign country/island is huge. You'll be able to attack off foes and then place a City right then and there. It's something that is not to be reckoned with. Highly Recommended.

  • Science
Chances are you will want to spread your cities out. Most of the time you will not be near Natural Wonders. When you are just read my Culture section and ignore science. As any Civilization if you are sprawled out chances are you will have a lot of science output because of the libraries (if you build them) and the like. There is no personality trait to help you outside of being near a Natural Wonder that gives you extra science. Recommended.

  • Culture
This is more of a situational victory. Reason being is that if you are not near a lot of culture giving Wonders then you do not really want to try this. On the other hand if you are near a lot of culture giving Wonders you'll want to create cities only near those specific Wonders and have a blast. Doing as such will make a Culture Victory somewhat easy - but it's all situational. Somewhat Recommended.

  • Diplomatic
Spain may or may not be nice in the real world. Ages ago they were not nice to Central American Natives. They are not nice in Civilization V. You should not be nice. The negatives of being nice highly outweigh that of the positives. There is no personality trait. Chances are you'll be spread out. That equates to some Civilizations that really can not stand you. Not Recommended.
Social Policies
Won't get into as depth as the Victory Conditions only because Social Policies can usually go anyway for any Civ and still win. Will only note those that are very important to each Civ.
  • Liberty
As any Civilization that favors expansion you'll want to pair up Liberty with you. It'll create many cities for you. In the mid game you'll be able to dominate by creating tons of Conquistadors and Tercios. Conquistadors will act as a Knight/Settler so they are very important to create in the event you are playing a Continents based map. Highly Recommended.
The rest are really up to you on how you're playstyle will be for policies, Tradition for hunkering down with few cities - Honor for cutting down enemies early on - Liberty for expanding like a mad-man but again, that goes for any Civ, not just the Spanish.
Factoids
  • Spain is traditionally a strongly religious country (Roman Catholicism). However, only 76% of Spaniards now identify themselves as Catholics, and only about 20% are regular church-goers. Due to recent immigration, 3% of the population is now Muslim.

  • Spanish-speaking cultures have been very propitious for the development of new dance styles, such as Flamenco (inspired by Andalusian, Islamic, Sephardic, and ♥♥♥♥♥ cultures), Merengue (Hispano-African), Salsa, Mambo and Cha-cha-cha (African and Cuban), Rumba (African, Amerindian and Spanish), etc.

  • Spanish culture greatly influenced modern art from the late 1800's, with artists like Antoni Gaudí (Art Nouveau), Pablo Picasso (expressionism, cubism, surrealism), Joan Miró (surrealism), or Salvador Dalí (surrealism).

  • 58 million tourists go to Spain every year, making it the fourth most visited country in the world. Spain's foreign tourist industry is worth approximately 40 billion euro, or about 5% of the GDP.

Don't really go too much into Pantheon selection due to the situational nature of things. If your looking for extra science choose Messenger of the Gods. If your looking for extra produtcion choose God of Craftsman. If your looking for faster growth go with Fertility Rites. Nonetheless, it's a matter of situational awareness. Which method of victory do you prefer?
