Leaving out the fanony-background fluff, this mission really speaks to me; especially if the fiends actually give you trouble because of the dialogue with the patrol you meet if they do.
Other than that, Dead Money.
After the fact, I looked up the quests on the wiki, just looking at the list and not descriptions and I'm only missing less than 20 sidequest and that includes about 5 you need to enter the fort to get which I haven't done yet.
So, more importantly, any must-see locations other than the vaults, which I'm picking off now.
There is a crater NE of the game, you can get a party hat so you might want to check it out. East side on the river there is a spot where you can get a set of Enclave Remnant Power Armour (Only the body). Just bring a bit more ammo than usual if you want to go there.
Also you might want to go Legendary creature hunting.

The only thing it would make it great, the one thing for which I would replay all of FNV now, is if the actual score for Auld Lang Syne played (bagpipes, of course) while the guys walk with you on Hoover Dam in their power armor.
Simply awesome.
And possible T-51b can be found there
