IIRC you can safely run forwards while dropping them as they take a second to arm. Either way the important thing is to run in a straight line while dropping them as the stupid AI will head straight for your current location no matter how many mines they step on.

No, it's not going to make any difference. Once they start coming after you, the frequency doesn't change just because you kill more of them. It's a fixed interval, and you also have pass certain points on the map for them to appear.
As for the NCR support, that's pretty much useless. I tried it once when I saw the assassins head my way, and by the time the single Trooper arrived the battle was already over. I never bothered with it again after that.
If your reputation with Caesar's Legion is Vilified, and the player is at a high level, assassins appear more frequently (about one group every day, attacking the player).
