I just finished the ascension 4 for the 3 characters and I was wondering : what are the 3 keys ? (emerald ruby saphir) can we finish a run without taking it ?

You can finish a run, yes, but you won't get the character ending / final boss without the 3.

Depends on if you want to do it. Allows you to see an ending, unlocks beta art, is a cool achievement, etc. If you don't want to, you can safely ignore the keys, but if you want to push yourself, you can try to beat the final boss.

Final boss is 250 extra score, you don't have to kill him (I got to him at 6th ascension, died, still unlocked 7th)

The final boss is way more than 250 extra score. It's 450 below ascension 20 and 500 on ascension 20, not including the ascension multiplier itself, because your bosses killed score goes from 300 to 500 or 500 to 750 on top of the 250 point bonus unique that boss.
