Asteroids do run out and don't regenerate/respawn if you mine them normally, if you mine them out, they're gone forever. The same counts for stations you (or another faction) destroys or pirate sectors you clear out. The whole galaxy is pretty static and dormant at the moment and not really sandbox just yet. There's no "do what you want, where you want, how you want" since the game constantly forces you to move on.
This will eventually change though, and you can expect to see the galaxy become more and more self-sustaining after the forthcoming alliances update. The out-of-sector loading addressed in that update will open up a lot of avenues for further dynamics to be added.
