At least I found no way of doing it. I just looked at the map, noted which lefts and rights I need to take, then follow the signs along the road and re-open map as required to make sure I'm on the right track.
I just tried it, and the only places that I can click on, on the map are cities that I've already been to. I was hoping to be able to click on a city on the map (even cities I've not been to) and plot a course for that city regardless if there were to be freight jobs there to pick up or not. Sort of like sight seeing the different cities. If it works otherwise, well... It's not doing what I'm trying to do.
The job market (starting point) is always limited to the cities you have discovered. The destination is not limited (obviously) so early in the game it's useful to pick undiscovered destinations.
If you own your truck it's useful to drive freeroam to nearby cities to discover them so that those jobs become available. So you don't need to make a job delivery to a city to discover it, just drive there in your own truck - freeroam with no trailer.
Suppose you've discovered London and Liverpool. You're currently in London or for that matter anywhere on the map. You can see there's undiscovered cities between where you are and Liverpool. Pick a job offer in Liverpool that's not necessarily a job that you really want. The GPS will route you past a lot of undiscovered cities, just drive into them to discover them on your way to Liverpool. That Liverpool job offer may expire but that's not a problem.
This also works if you're actually on a job, drop your trailer (T key) somewhere safe and take a moment to drive your cab into an undiscovered city that you pass. Go pick up your trailer after discovering that city and continue on your way. The only issue here is that these extra visits may make you late. You can drive thru cities cab trailer and all if you like, it's sometimes hard and time consuming to get turned around and back out of the city with the trailer attached.
I doubt they'd change it now but it sure would be nice if we could select any city (discovered or undiscovered) and choose to have the GPS plot the course to get there. To me it's far too tedius to have to keep consulting the map to see where I might have to make a turn to get to where I want to go especially annoying when there is a GPS unit sitting on my dash which unfortunately only slightly simulates a real world driving GPS unit.
