If you are not on Windows or your GPU doesn't support DX11.1 then you should use OpenGL.
You should generally use 64bit as this gives the game more RAM to use.
If you just select "Start ETS2" then Steam will use the option you chose for the last successfull launch.
Typically you'd only need to select DirectX 11 64bit once(on most systems) and you'll be set. OpenGL is an alternative if you have some issues or are playing a non-Windows operating system.
