Euro Truck Simulator 2
Route Planner Navigation Mode
In the "Gameplay" settings section, there is an option for "Navigation mode" and it has three options, Best, Shortest, and Small Roads Only.

Best seems to find the fastest route (in game time), Shortest finds the shortest in terms of actual distance, and Small Roads Only is pretty self-explanatory.

My question is, what's the benefit to driving the shortest route? It's longer in game than "Best" but is it faster in terms of real-life time?

Thanks.

My guess is that "shortest" means you drive less km and need less fuel.

It is not true. Imagine situation, when you are driving throught 5 cities, every city has a ring road. Ring road, is longer, than driving throught city center... Shortest way will give you longer time, and hgher fuel usage - because you will have to stop and start rolling again several times.

Best is considered best. It has longer distance sometime, but it should be on motorways, so you will have better time, and better fuel usage.
What is benefit of using shortest? I guess nothing. The same with only small roads... It will just give you alternate route, you might want to see, you've never been.
I always check the route, and alter it to my needs anyway. Sometimes I find better, more funy, or more complicated, or the one I've never driven before. After picking up a load, I'm checking map and planing route myself. Automatic navigation is a sugestion that I'm not always agreed with.

The irony is that I was about to write the same – I never use the shortest route because in the end all the intersections, turns and roundabouts take longer than using the highway – but then I didn't because the OP asked for the benefit of the shortest route.
I have mine set to small roads. The amount of time it takes or fuel used isn't relevant to me.
Best = Most direct - often the fastest too.
Shortest = Least amount of miles - but maybe slower due to speed limits, junctions, and terrain.
My advice would be to stick with Best. I had it set on Shortest and got stuck under a low overpass. With a Yacht on the trailer.... no joke.
