Use your shotgun for close range to get the most damage out of it, if it's a rifle you can use it for long range instead.
Revolvers are pretty versatile, when your close range it's pretty hard to miss unless you're going for the head, stealth hits at mid and far range are great, especially when you get better revolvers.
Knife is pretty self explanatory, I usually don't get too many perks for melee but grab a couple if you like the role-play side.
Personally I can't stand explosives, they're way too clunky in 3/NV but if you use vats and invest in the right perks, they can be used well at mid-range, crowd control is sorely needed when all your guns are only good at single-target elimination.
I love my Cowboy playthroughs... revolvers (get the mod for "Clint's stuff" (Eastwood that is) and get the revolver "Clint's Handful"... I have also found the repeaters with the upgrades you can buy from Vendortron throughout the game at Gun Runners does that job well at mid to long range. You can get scopes for 44's - not really cowboy but that's where VATs is your friend.
For "Cowboy" style sniper work get La Paciencia or the Trail carbine with scope.
For the clothes hit up the mods for the Desperado Clothes and use the Gray Ranger Hat with the gray to black conversion mod from Nexus.
I am currently playing through with six companions 'n' guess what... we're the Magnificent Seven....

P.S. If ya wanna go "perfectly Cowboy" then restrict yourself to the 357 calibre guns, about as close to a six shooter as you'll get unless there's a mod at nexus for a 5.56 calibre revolver. The Ranger Sequoia is also a good bet.
There's also the Weapon Retexture Project, and it's hotfix, which will give all the guns in New Vegas a sexy makeover. Make sure you get your load order right though, otherwise the textures will break. You want the order to be;
