As leon you need to open the club door in licker hallway of death.
Once you make it in the Club room, Mr. X speeds to your position.
Running out of the clubroom into the licker hallway of death is a bday day so you walk over to the Heart Door (that leon can't open) and try to open it. You will twist the door nob and for what ever reason it despawns the lickers in the previous hallway.
Yeah, that one doorway they camp...Love it. You just have to keep your sub weapons for these moments. That is in my opinion the most resourceful strategy to get through s+, which is not missing or wasting any sub weapons. They are basically "get out of jail free cards" for any situation where you feel overwhelmed. You can easily spend an extra 10-15 minutes clearing the police station properly if you want (including looting the whole weapon storage room) and still make the 2:30 mark. This will make the rest of the game much easier. Part of a speed run strategy is having sub weapons that can help you bypass these rng moments at the least, until you have memorized how to deal with each room perfectly.
For a first S+ run some good save points are the Club Key, Sewer Entrance, Lab Entrance. Once everything becomes second nature you can switch the last two to before G2 and before G3. G1 imo isn't worth a save.
It is a pure guess, but I think just like in RE7, the door opening screen actually puts you into a hidden small area way below the map for that one camera shot (RE7 used this for the shadow puzzles) to ensure no enemy attacks you during it. This should put you in a spot where enemies in previous rooms are reset to their spawn points.
You can kill them easily with the knife, or if you want to go pow pow with the magnum.
Of course, it is not advised if there are two of them in the same room or Mr X is near.
