Unlocks meaning that you can 'unlock' better things that will help you to progress in the game.
And it's been awhile but I don't think you can just press it to do anything? You actually have to earn the points through play throughs to unlock the awards.
Rogue likes means you don't get to try the fight again. Once you're death you have to start back from the beginning.
This may sound very punishing at first, but it actually is really addictive once you get into the gameplay loop
Also, while I don't recommend this, you can savescum by quitting the fight before your death. By doing so you can start the fight over.
The unlock button is to unlock more relics and cards that will show up in future runs. As you get more experience you'll unlock more of them.
Click the unlock button, enjoy your new stuff, then start a new run from the title screen.
