The Daily Climbs offer runs with various modifiers that shake up the core gameplay a bit. There's a new daily every day to check out.
That said, this is a roguelike, and usually people can play the base game of a roguelike over and over because the bulk of the game is randomized, and the core gameplay is fun enough that people just want to keep playing the game. That's been the case for me, at least. I bounce between retrying the highest Ascension difficulty and doing winstreaks on the base difficulty.
Yeah, you can either beat each boss at least once, or beat the game a total of five times.

Aside from that, one can replay and attempt to win in different ways, and multiple attempts will be needed for some of the harder achievements (e.g. you can't really force a Minimalist deck win, but if you get a very early Peace Pipe you might think about going for it).
I want to ask about deck size , how many cards should i go for in a decent deck
It's more about each individual card selection. It's thinking less about "is this a good card" and more about "would adding this card make my deck better".

However if you can't differ between good cards and cards that make your deck better, the deckbuilding for dummies recommended size is something like 18-24 cards. Exhaust/powers counts as 0.5, offering/adrenaline doesn't count at all. And going lower is always better.
Hunt and grind to get all achievements,
Achieve ascention 15 with each character
try to get in the top 100 in a daily 12+ hours after daily changes.
Set your own goals. try to make an unconventional deck work. try to get a single card to do over 2000 damage?
