Another reason it is highly replayable is because each character is totally unique from each other (they can only use their own class cards, whatever you call them). Therefore you will need to play the game totally different with each character to have a chance to win. There are three characters total in the game (2 characters playable, and 1 character playable in beta if you choose to play the beta version of the game).
Yes, you must start over literally from scratch when you die. I would call it a rogue-lite because you get unlocks similar to Gungeon. Exactly how Gungeon unlocks new items (they don't give it to you. It just means they are possible to gain from the random loot chests or monster drops now.) I'd say Gungeon is rogue-lite too. I have never played The Binding of Isaac.
I'm not sure what you mean since I've never played Binding of Isaac... The normal game stops after you beat the final boss (currently). There is additional daily challenges and one of them had a infinite floor mode. Meaning, you are playing forever... After you die, then your score is added to the leaderboards to compare to people in your friends, region, or the world. It's very competitive, and some of my scores place as low as 45,000th place... So, there are a lot of people playing.
The game play is closer to Hearthstone with only 3 classes and perma death, but with a slightly lower quality of sound and graphics quality (You can view it on gameplay videos easily).

3 levels, 15 floors each, up to 13 of which give you a choice of 3 cards (excluding events).
3 bosses, 1 from a pool of 3 per level, with special rewards after.

Yes, it is endlessly replayable with variability between games. Plenty of us have more than 100 hours of play just from the first two characters. You will get your money's worth from Slay the Spire.
