Cripple a leg and they will start to limp. They can only move very slowly and will drag their leg behind them.
Cripple an arm and they will lose accuracy, but can still shoot a gun if they have one. They are very likely to drop their weapon, but might pick it back up if they live long enough.
This is all humans mind you. Other creatures might react differently. Feral ghouls will actually lose the limb you target and keep fighting, even if they have to drag themselves forwards with their hands.
I've played a lot of this game and the Fallouts before it but I've never seen an enemy actually lose their weapon in Fallout 4. You can't even shoot guns out of enemy's hands, which is a bummer as it was really useful in the older games. It seems that enemies getting disarmed has been sectioned off to the Gunslinger perk.
