The great thing about rimworld is that you never really lose. If all your colonists die, then you just wait for about 30 seconds and then a wanderer joins and finds the ruins, and you can start again. Although in some cases, you may just want to quit.
So alter the question to say how long does it take for all colonists to die out, then the answer is that it depends on the difficulty and your skill. I usually play for around 10 hours before the raids get big, and moods get low. But I'm getting better

Ooooh yea, Dis game has no tutorial so your gonna start over a few times anyhow. Other then that other areas give different tactics you can use, also with multiple story tellers and a bunch of mods you can replay this almost infinitly
Don't dabble too long over a lost colony, the retrying again and again is a big part and a big charme about the game.
That said there is already a ton of mods that add complexity and longlivity to the game. I'm not sure if they are up to date with the current steam version but it should be around the corner I would think.
