I didn't actually do the beta version thing, but I did have to go to the audio options and tweak it to shuffle, otherwise it'd play the vanilla soundtrack first.

You have to update to beta version: Right-Click on games, propriety, go the the Beta tab and change none to the twopointhospital_patch_beta let it download again and play.
