I only go to the mackeral spot to stock up on Emporor's for the commisions from Django.
Mmm, I tried both and found the waterfall more profitable, but at the time I wasn't using a horse so It was taking a while to get to the mackeral spot which seriously dented my time available to fish there. Maybe with a horse it makes a big difference.

Yeah you can breed fish. I've found it to be a real pain in the butt though persnonally. For a start Goliaths only eat Bug Eggs which is a pain to get as only LadyBugs drop them. Many of the other types are happy with Dough that you can make or get from Sophies farm but those 2 types certainly won't eat it and die if you try.
Second you have to feed them everyday, make sure there is space in the tank for the "baby" and give them just the right amount of food (and that doesn't seem to collate perfectly with the amount of fish in the tank so ii's hard to judge).
I tried starting a fish farm with 4 Large fish Tanks and although I got them to breed on the odd occasion it was few and far bewteen for all the bother involved.
Perhaps if they change it so your helper feeds them and the amount of food needed is consistant then maybe it will be worth it in the future but IMO you are better of just fishing for them at the moment.

As you can see, waterfall and desert river are essentially the same, but the waterfall is much closer.
