How do I donate fish to the aquarium?

When I go to the research centre and try going upstairs, there's no "enter door" prompt or anything at the door, it's just blocked off?

If you stand in front of the aquarium, very close to the glass and select the fish in your hotbar, you'll get the option to put it in the tank. Almost the same mechanics as when adding something to the cooking set.

The door at the top of the stairs in the research centre is one of the many doors in Portia where stuff could have been but hasn't been added (yet?).

The aquarium is in the museum, not the research center. Go into the museum, up the stairs, and then do as JVC said.

Also, you only need to donate the Kings to get all rewards.

There is an inside door from the research center into the museum so it's a connected building.

True, but the OP was trying to reach the second floor of the museum by going to the top of the stairs in the research center, which has a door there that doesn't open.
