As you fight monsters the locales level up. This can allow certain monsters to spawn, or make monsters tempered if the level is high enough.
There are also minerals to mine and bonepiles to gather from. These can level up as well to provide more rare resources, and eventually spawn a large node on the map that is guaranteed to give the rarest item once, which resets the resource level.
You can only gain so many total levels for all of the zones put together, so only level up the ones you need. You can use the handler to downgrade a locale if you don't want it to level up, or even make it so they won't change at all. As your MR reaches 49, 69, and 99, you'll get assignments to upgrade the max level you can get your locales to.
You will also unlock lures by finding "signs of turf wars", and hunting monsters. These are a consumable used at the handler to force a monster to spawn where you want it. You can also unlock the ability to meld lures once you encounter the respective monster in the guiding lands
I suggest hunting monsters in the wildspire/Forest region and (once you unlock it) coral region.
That's my personal way. And at the end I ignore/replace (once I get enough gold rathian mats) wildspire and go for coral and volcanic.
But you most likely have yet to unlock those two regions so just keep leveling up one of them up to 4 by hunting monsters in there.
The most tedious and annoying thing you do there is "farming" resources by going in cycles in a specific region and picking up bones/ores (they have fixed spawn locations but random spawn timings). If you take a look at the map overlay, while in there, you can press a button and change the guiding land info to a "resource" related information. There is a bar, you fill it up by farming resources and when the bar gets filled you will start to find more rare/exotic items.
Important stuff to note:
(All monsters here are tempered variants)
Wildspire 7 - gold rathian
Coral 7 - silver rathalos and namielle
Volcanic 7 - rajang (appears in many places but volcanic one seems to be the easiest one due the layout of the area), seething bazel, Kushala, Teostra, Lunastra, Ruiner Nergigante
Honorable mention:
Forest 3 - Garuga (one of the best greaves/legs or whatever you want to call them, in the game): Garuga Graves ß+
edit: many regions are required for specific items to upgrade/augment weapons at the highest ranks
There is a total level cap, so you will never have everything maxed out at the same time. Force 3 regions down to lvl 1, then push the rest to lvl 7. Tier 7 is super important and most of the guiding land rewards are there.
There's nothing too special about leveling up a region, just keep hunting monsters that spawn in those areas.
Pinned guide of what to do in GL.
btw most high tier part are ONLY available in the guiding lands
theres no quest.
you just wage a war vs the monsters. breaking parts is very efficient. just kill monsters.
slaying/gathering in an 'area' levels that 'area' up, and then stronger monsters/better materials may appear from that area.
guiding lands drops materials only found in guiding lands - you won't find it anywhere else in the game.
if you want to get the r10 and higher stuff, you have to do a lot of guiding lands. you will need to cycle every area to rank 7, and fight its strongest host of monsters and gather the best stuff from them.
Of course you do have quests that you need to do to unlock content :
- To the Very Ends with You (you need MR99) as an assigned quest.
- Reveal Thyself, Destroyer (MR24)
- Across the Lost Path (MR24)
- ...And My Rage for All (MR24)
The last three are special assignments.
- Achy Brachy Heart (MR 24) as an optional quest.
