Wait for the creeps to lower the opposing teams creeps. Hit the creep when it's nearly dead. You can also turn off auto-attack in the settings to get better chances on last hits.
You can start by playing Limited Heros game mode. It has all the heros that are used for beginners.

2. On the main menu click the little gear in the upper left corner which will bring you to all the options starting on the hotkey screen. Find the action you want to change (mouse over for additional information) and hit the button you'd like to change it to. In the upper part of the left section of the hotkey tab is your spells and just under those buttons you should see some labels, one of which should be quick cast. Click that then any buttons you assign to your spells while in quick cast will cast your spells wherever the mouse is at the time you press the button.
3. Some heroes have higher base damage than others and larger damage spread which can make last hitting difficut. The creeps have a rhythm as they attack the other creeps and its different depending on how many creeps are attacking and when they began attacking. You'll get better at finding this rhythm with time but it's best to attack slightly before you expect the creep to be within last hitting range. Attacks take time especially from ranged heroes who have projectiles so begin your attacks slightly earlier.
4. It all depends on who you prefer playing in these types of games ranging from spell casters to just right clickers, supports or carries and all that. But some of the easiest heroes are often the right clickers and carries although their are some easy casters (called nukers in dota) and supports like:
Lich:Nuker/support, very easy to understand and use spell
Wraith King:carry/attacker, arguably one of the easiest heroes in the game with only one active spell to use which is a stun and chase spell. His other spells include lifesteal, a critcal strike, and the ability to come back to life.
Bane:nuker/support A very strong disabler with good damage but does have the weakness of focusing multiple people ate on time.
Warlock:nuker/support A healer and an amazing teamfighter with his spells being so powerful however he is very ultimate based and requires some farm.
Vengeful Spirit:Versitile She has good spells for chasing and increasing her team's effectiveness but can also use this for herself and can deal a lot of attacking damage as well.
Sniper:attacker/carry Requires farm and has low health but a very long range and he scales well into the game.
5. If you look near the towers in the shortest lanes on the sides you should see pathways leading to the secret shops.
6. There are two kinds of wards: observer wards and sentry wards. Any other types of wards are spells from heroes that have different uses. These two wards can be bought from the consumable section o the main shop and the observer wards provide vision in a large aoe while sentry wards will reveal invisible units in a large aoe. Oberserve wards are yellow and cost 75 gold each, last 7 minutes, and come in limited numbers that resupply over time. Sentry wards are blue cost 200 for 2 wards, last 4 minutes, but are always available.
7. In the lower left corner of the screen just above the fortification glyph there should be a red "X". Just above that "X" is is an item called town portal scrolls that let you teleport to allied structures anyway on the map after a channeling time. They can be bought from the main shop or the side shop in the trees nearby to where the creeps first meet in the side lanes. They have a lengthy cooldown of 70 seconds, cost 75 mana, and have a channel time of 3 seconds if you are the first to teleport to the allied structure after a time or if you are teleporting to the fountain. If you are not the first to teleport to a structure after a time you will have a 5 second channel time. Carrying a TP scroll is very important for defending towers and escaping some surprises from enemies or setting up your own.
