I was just about to play Dota and I get this message:

Dota 2 error reading from loaded packed store pak01_001.vpk.

Have you had this before?

1) delete those files that cause the error

then

2) verify integrity of game files

it should fix the problem.

I tried this. Deleted two files that were causing error. Verified the integrity of the game files and when i launched it. It showed two different files that had error. Cant keep doing this forever. Is there any other fix?

the fk was that..... i was abandoned twice because of that

Uninstall all the pak01_nnn.vpk file from the folder, update your client via Steam and verify integrity. It fixed mine
