Should I play as Duke on my first playthrough?

I have not played this game before. Should I play with default protagonist to get the original experience, or can I ignore it and play as Duke from the start?

Definitely not. In fact I'd recommend against buying the Duke DLC in the first place .-.

why? Just because of consistency/quality or the DLC can cause some technical issues?

I have played the original and recently got Full Clip Edition. Played through it as normal. Just recently decided to try the Duke run through. 5 minutes in and I wish I hadn't. Duke's lines are basically Grayson's redone in Duke's voice. And Duke has not aged well. I've never wanted to skip cutscene before.

Don't play as duke the lines he makes make no sense at times and the chars still address you as grey. it's like they did a brain swap where everyone but you see's and hears grey and you see duke.

That would require time and effort and the publisher wanted to pump out the remaster for a quick buck.

Only play as Duke if you replay the game.

On a first playthrough, just play normally.
