You have to have taken the "Showdown" sidequest which requires the bank job and train explosion sidequests to unlock. If you haven't gotten showdown, she won't spawn.

There is the bank quest and the 3:10 quest from the train station (if you can't find it, restart the game while in lynchwood. It is close to the fast travel station).

You need to finish BOTH of them. Then Showdown is unlocked. Take that quest and the Sheriff will spawn.
