Collecting all of the rare, unique, legendary gear?
All missions, all DLC content, all headhunters?
Or do you mean finishing the main story? Once.

Well, the DLC's can be done in the first playthrough, but are meant to be done in the later ones. There are some enemy scaling issues where you can find certain DLC enemies are level 50, even in Normal mode. Needless to say, you will be nowhere near that and they will be essentially invulnerable. The first playthrough (Normal mode) is capped at 30 - 35, so gear will not be better than that.

The time to play through it all will depend on several things, knowledge of the game, luck with gear drops, your game skills...

I can complete any of the DLC's in 4-6 hours with a new character. All missions. I have done it many times before and know the where and how. Doing it yourself for the first time when you do not have that knowledge will take rather longer.

There is one mission called 'You. Will. Die. Seriously'. This is given out at the end of the main story after you have defeated the final boss. This mission is never lower than level 50, and the boss level 52. Basically you can't take this boss on until you are that level, which in turn means you need to get to level 50, which is done by completing the 2nd playthrough and leveling up.
You could get a higher level character to kill the boss for you, but then it wouldn't be you doing it for 100%.

There is a lot more to this game than doing the missions with one character. Why the rush? It is the journey that is fun, and with this game there can be so much more than simply completing missions.

all missions plus DLC mission on NVHM with a fresh character can be done in around 20 hours probably

exclusing the level 50 requirement mission since for that you gotta just level up fo another 20 hours

if u want to do 100% i would say about 1000h it depends if u get help or not and how lucky u are
i have to add that it depends how experienced u are with the the game too

I could link you these 2 pages on the official speedrun forum, if it does help you in anyway since iirc borderlands 2 is usually run with a gaige anarchist skillset and on an older version where you could do an infinite shooting shotgun glitch and ofc people exactly know where to go and what to do and all that.

The BEST way I have found to answer that question is, play the game and find out. The answer is vastly different to different people. I would think it is complete when you decide not to play anymore.

My first play of the story was a LOT longer than what some others had, I snooped about exploring all the out of the way places and tried to get to places that may not be intended, found some cool stuff too. Others are only interested in how fast they can get to the end, I never understood that but to each their own.
