You can, at any time, go back to the menu and switch to a difficulty you have unlocked already.
There are 3 difficulties (Normal, True and Ultimate Vault hunter mode - nvhm, tvhm, uvhm) and for uvhm, if you have the lilith-DLC; there are additionally 10 OP-levels - unlockable extra-levelups guarded by a ♥♥♥♥♥♥♥ of monsters and some strong bosses.
The story-progress is saved for each difficulty individually. A mission finished in tvhm is only finihsed there and will not affect uvhm for example. So when you start tvhm then you can do all missions again. If you finish it in uvhm but havent played it in nvhm the quest is still available in nvhm.
Aside from that - in tvhm you want to make sure to NOT be much higher than lvl 50 when finishing it as in Uvhm enemy-level scales with your level, but in tvhm you "only" get equip up to level 50 - if you level higher the mobs will shredd you to pieces.
...but this will only hit you after TVHM. So don't worry too much about that right now.
You just want to avoidthat.
Also depends on who you're playing. I'm pretty sure Siren with Ruin and Converge in Motion will breeze through it.
As other people have already posted, yes you can. It will also save any quest only items like the grog nozzle to the mode, so if you got the grog in normal mode and still on that particular mission, you wont see the grog in your inventory when you switch to tvhm (or uvhm), but will see it again when you switch back to normal mode.
But why not just do the DLCs that you haven't done in tvhm instead?
This is pretty useful on saving time. Example is there is a particular raid boss where I have to go through a particular area that is a pain for me just to get to the nearest travel station in UVHM, so I just switch to TVHM since I haven't done that DLC in that mode, and takes me to the travel station.
...retry in UVHM to get your Lvl 5x lazy eight. There the maps (and the baddies' level) will always be synched to your level.
