At most you'll be able to reach capstones on two trees.
In addition, some skills in a given tree require that the preceding "level" has had a suitable number of points placed into it.
Which skills can't you "buy"?
If it something like that http://bl2skills.com/assassin.html#505051045150055145015050500000 (for example), you can't spec into skills you wanted anyway. 'Cuz it's max lv.
And overpower levels don't give you more skill points.

I honestly can't figure out what you've written here other than to say that you've indicated that you know how to "buy" skill points.
Until you can post your current build as CommanderDouchebag asked earlier so we can know where you are starting from with this question, my earlier response in post #2 still stands.
In order to start to put points into Zero's Backstab in the third tier of his Bloodshed tree, for instance, you'd first need to use ten points:
1. Place at least five points between Killing Blow/Iron Hand in the first tier; and
2. Place at least five points between Grim/Be Like Water in the second tier.
Once you've spent these first ten points, the third tier of the Bloodshed tree opens up, including the Backstab skill.
If you haven't done both 1 and 2 above, you won't be able to put points into any third tier skills.
Could this be what is occuring? Otherwise, I'm at a loss as to why your "clicks" aren't registering.
