You can't force the Black Queen to spawn but there is also no need to farm her.
The XP relic is a complete waste of a valuable gear slot, you shouldn't really bother with it. Use relics that enhance your characters abilities or balance out some shortcomings to maximize your efficiency in combat.
The XP gained from killing enemies and completing (side)quests are more than enough to get you through the game, especially if you play it your way and insist on farming all the time.

Given how long it would take to drive round and check all 5 locactions, it might be quicker to save, exit, restart, check the box in Ellie's garage until it spawns.

1 Inside Ellie's workshop - When entering through the vehicle bay nearest the fast travel, in the corner next to the entrance.
2 The Dust, East Side - On the West side of a rock outcropping, South and slightly East of Ellie's Garage, North and slightly East of the entrance to the Hodunk Speedway.
3 Goose's Roost - Inside the shack at the lower end of the stairs along the North side.
4 Lynchwood station - Located on the Northeast edge of the platform close to the New-U station.
5 Old Dahlwell Oasis - Next to a power pole due North of the gas station building containing the slot machine.
The Black Queen occasionally bothers to turn up and occasionally will drop the Nukem launcher. Occasionally * occasionally means Nukem drops are kind of rare.

With the "Clan Wars" string of sidequests completed, I'd check the latter two to get a shot at the Maggie or Slagga aswell.
She drops the Nukem sometimes when killed, so it is not completely worthless to farm her.
The Hyperion barrel provides an additional critical hit damage modifier, the special Gwens Head variant grants you even more and it is multiplicative.
Compared to a blue Dahl pistol with the same parts, it's stats are a lot better and it has a higher burst count:
