Ahh... I remember my first Buffalo. I took that thing to Sawtooth Cauldron and had a blast with it.
Lets say it's a mission boss: dukinos mom, she can drop a legendary launcher called the mongol, she also has a set of rare skins.
The way you can farm her is kill her for the mission, go back to where you got the mission (the sanctuary bounty board I believe), and you turn it in, then you save and quit. You load back into the game, you go to lynchwood, and you go down into dukinos cave, you kill her again, she doesn't drop anything good, you just save and quit, go back to the cave, save and quit, blah blah blah. Some people say certain bosses will respawn when you travel to another location and back, but I haven't found that the case with everything.
In summary: turn the mission in,(some people you want to farm for aren't involved in a mission, like savage lee, he just spawns in the world, and you just kill him, and keep saving and quitting to get one of the best guns in the game) save+quit, go to area again. Boom, that's how you farm.
