There are 2 more playthrough modes, not to mention the DLC's, so your looking at a lot of hours on top of what I mentioned.
If your a perfectionist and really enjoy the game, then as stated above hundreds of hours.
Yes, each chracter plays differently enough and each has their own strengths and weaknesses.
This game has a lot of mechanical content, and the game world is also vast. You will get 100s if not 1000s of hours out of it.
I didn't bother with the OP levels with her, although I might do so in future.
I also took a completionist approach to the BAR challenges. Every challenge, every vault symbol etc completed apart from 2. These two unfinished ones are Jimmy Jenkins (only found him 4 or 5 times in 3 playthroughs) and the revive a team member (only level 2) - basically I very rarely play co-op and when I do it is only with friends, so this will take a while to complete. Everything else, maxed, done, completed and stuff.
This character took just over 200 hours in total.
OP. There are 5 other characters, and yes, they all do things differently. This game does have a lot to it, and Pandora is just a nice place to wander around, occasionally shooting enemies in the face.

We need to start by defining what exactly we're measuring with the description "the game" - to me, the game represents taking one class from lvl 1 upto lvl 80 / OP8 using the main game and all the playable-quest dlcs --- optional missions are not required. When done using no third-party-software / no glitches / no nonsense, your first game will take about 300 to 400 hours. Your second game trying a different class will shave +/- 25% off the time it took you on your first game. By your sixth game, people get this down to +/- 200 hours.
By their twenty-fifth game, experts can complete the MAIN GAME ONLY (all main quests in BL2 no side quests / dlcs required) and bring this down to +/- 3 hours. I believe some people call this 20-mission basic game a "speed test" aka "timed test" though I can't be sure what it's called cos I really couldn't care less about speeding thru the game - it's not an attractive way of playing BL2 to me.
When people ask how long it takes to complete the game, they mean the main story :P.
The main story is a fraction of the content and challenge in the game, so no. How long is the game? You haven't "completed" the game until you have solod one character to OP8,doing all raids and circles at OP8, including farming all the necessary gear. No dupes or Gibbing stuff in. This way, the game is 300 or more hours long, per character, easily.
Doing EVERYTHING would take 200+ hours
