Lohse has a nice side story, Fane adds insight to the main story and has a great support ability, Sebille is an elf, Ifan gets a leg up during act 1 (a level 5 crossbow that deals as much damage as a level 7 weapon and some extra xp at the end of act 1).

It's not a big deal.
Fane is good because he has a very strong origin ability that is useful throughout the game, and scholar and mystic tags which are the only useful tags. But you can get access to both of those if you bring him to your party. He's also tied in some with the main story.
In comparison, Ifan's origin ability isn't very good, you probably won't ever use it once you get other source spells. His tags, soldier and outlaw, don't give any really helpful dialogue options. His story does give you a little background on what happened just prior to the game.
I don't get the Lohse recommendation unless you really like deals with the devil. Her origin ability isn't great. She does have mystic at least, and she's kind of useful in that you can get the entire party's collars off as soon as you escape fort joy (beast can also do this). It might be because she's not a murderhobo like Sebille and the only other female option. She's a complete side story though.
Honestly, though it doesn't make a big deal. Especially if you plan to playthrough more than once as you can do 4 of them in one run and the other 2 in another run. You're not going to be on hard mode because you picked Ifan or get on easy mode by picking Fane.

As a new player, I'm doing a 2-man run (Lone-Wolf) with Lohse and Sebille. If I complete the game and feel like doing another run, I'll use the other four characters and treat them like extra content (seeing as the main reason for having any of them is mostly their fully fleshed stories + unique skills).

No matter who you choose - even if you go with a custom character - you won't be crippled, so go with your gut and take whomever you like. If you enjoy the experience, you can play around with the other characters and explore their quests and stories to spice things up.

why is mythic and schoolar so important?

Mythic+Scholar tag combination gives unique dialogue options and ways to solve a few quests. It's the only combination that requires a character to have both in order to get them and Fane is the only origin character with this combination.
All other dialogue options require only 1 tag (at least to my knowledge).
Sill it's no big deal, there is usually another way to complete these quests if you don't have this tag combination and if you are really stuck and don't want to have spoilers by using a walkthrough ask people in the forum.

I didn't say they are important, just that they are the only useful ones. You can get by without them. Some objects in the world require one or the other to meaningfully interact with, including reading some books. For example, there is a quest that involves a recipe that you can't read if you don't have both on the same character. The quest can still be solved without it but you get lesser rewards unless you look up the recipe somewhere. This isn't a very common occurence though, like once per act maybe.
