John Wayne was basically an exaggerated symbol of masculinity for a whole generation of American men. Not much more to it than that.

The film The Green Berets was much loved (and ridiculed for its over the top acting and unrealistic plot) by conventional US soldiers and special forces alike. The song “Ballad of the Green Berets” (featured in the movie) was written and performed by Barry Sadler, a Green Beret who was wounded in Vietnam by a punji stake trap.

Popular American actor, he did a lot of cowboy like movies too. His real name was Marion Mitchell Morrison, bit of a hard name to remember so he went by John Wayne professionally, or Duke as his nickname. Very popular in the 50s, 60s, and so on, and I guess a lot of the troops had seen his movies. Not exactly sure why his name is used in a taunt, but there you have it.
