IMHO once you get the Strong Back level 4 perk, Supply Lines don't matter anymore (unless playing Survival mode).

Edit: and if you are willing to micromanage like me, Supply Lines don't matter before then either.

In thinking this over somewhat, I think Graygarden makes for a suitable central distribution center as long as I don't invite any settlers to it. The robots are pretty capable of defending it while having a decent turret defense along with a few guard dogs helps keep it pretty secure.
You can supply power there by placing the source on the top roadway. There is just enough room to add a few more food crop items to max the output to 36.
(I usually add razorgrain since that is the one food item they don't usually grow).
It suspect it also makes for a good material distribution center that can be shared with other settlements as needed. (just bring junk, weapons and armor to your crafting stations there and do what you will with them).

From there, you can distribute "fresh food crops" to other starting settlements by tactically sending caravans from the other settlements to Graygarden. This way, the population remains consistent and happiness stays pretty much unaffected.
Hmm, are there any settlements that let you crush your enemies, see them driven before you, and hear the lamentations of their women?

Hangman's Alley.
I like Red Rocket Truckstop. I setup my trade lanes like this, to minimize my guys moving through enemy encampments:

I don't think there is a best settlement, if you had asked what is the safe'st settlement you may have gotten different replies. There is too many variables eg a fair few get crossed off the list as they can be attacked from within, others are out on a limb and not close to anything, so that narrows it down a fair bit, wherever you chose just don't put any settlers in it period.

Nowhere feels safe.. I like my diamond city home most. Sadly it's disconnected from settlement network.

Hangman's Alley.
Hangman's Ally is a miserable little rat hole. I hate that place. Barely able to build a sheltered bed. It is good to dump loot... i mean, scrap, tho'. Better than Home Base which is behind TWO wait screens, HA isn't behind any. I think Bethesda went out of their way to make it difficult to build there. You can't even go "up" because it's only a few floors high.

The castle is a cool settlement, get a kick out of restoring the walls and turning into a proper fort.
