Sometimes just moving to a different spot close to them on the map pops things out. You usually get hit once or twice though.
If that fails walk off the map far enough that it disappears and come back.
If that fails try reloading a save.
Either try loading your last save and going there again or just leave the area and return.
Alternatively, find a bed or a sleeping bag and take a 1h nap. This usually gets NPCs unstuck as well.
Keep in mind though that this quest bugs out easily in the beginning. It's not unusual to see the quest marker but the mirelurk is nowhere to be found. You'll see this in other quests as well, especially the "Cleansing the Commonwealth" quest that Knight Rhys give you.
As for the missing mirelurk, sometimes the only way to fix it is to load an earlier save and try it again. From my own experience with this quest, I'd recommend loading a save from just BEFORE you discover Salem and then go directly to the Rook House. It seems to bug out less that way.
quest done...
Additionally, the third also never spawned, so I am essentially barred from seeing what else could happen. This probably happened since I showed up after the main questline and 87 hours in a single save. Unfortunate to say the least.
