I have this same problem, but mine is not going away. I've tried starting the game about 6 or 7 times in the last ~20 hours, to no avail. I don't use controllers at all - just mouse and keyboard.

The default is controller on, which disables the mouse.

Check it's off in settings.

(a) I can't access Settings because I can't click on anything
(b) The mouse is not disabled. The cursor moves around, and I can highlight options with both the mouse and the keyboard. It just refuses to acknowledge clicks and pressing the enter key.

You could try this........

Go to: C:\Users\Username\Documents\my games\Fallout4 (Or wherever your Fallout .ini is) and open fallout4prefs.ini . Find bGamepadEnable and set it to 0 and save. (It could be set to 1 gamepad enabled).

Yeah, I already tried that as well. I spent some time looking online to see if there were any fixes, but I've come up empty. And Bethesda's tech support line never called me back when I left a message. I seriously am pissed about this - the patch appears to have @#%^ed my game, and no one is able to un^#&* it.

Lmao. Here is what I wrote in to Bethesda:

"Hello. Prior to the most recent patch, I was able to play Fallout 4 with no issues. After downloading the recent update, I can no longer play the game. When I start the game, everything happens normally until I reach the main menu (Continue, New game, etc). At that point, the game stops recognizing clicks from my mouse and usage of my Enter key. Neither the mouse nor the keyboard is disabled - the cursor still tracks my mouse movements, and I can continue to use certain keys on the keyboard (such as the tilde key). I do not use a control pad, nor have I ever plugged anything other than my mouse and printer into my laptop. I would greatly appreciate your assistance in resolving this issue."

And here is their reply:
"Thank you for contacting us, I will be happy to assist you.

You will not be able to change the key bindings for some actions in Fallout 4. This is working as intended."

.....#facepalm

I'm having the exact same issue! It's frustrating because no one seems to understand that the mouse works in terms of highlighting things, but won't let me click at all! I've tried with a controller and the same problem. It's almost like the menu screen isn't linked to the screens that should be prompted when you click on a menu item.
I will subscribe to this forum and update if I can figure it out..

I was able to resolve it by using Steam's diagnostic tool.

Right click on the game in your library --> Properties --> Local files --> Verify integrity of game cache.

It basically said "Two of your game files are jacked up. We'll replace them with un*&^%ed versions." Once it did that, the game worked.

Thank you, it worked for me. I've spent the majority of my time after installing the game trying to figure out how to get it to work.

Besides any other fix mentioned here, to those still having problems with this, and specifically use teamspeak, the is an overlay and app named Overwolf. While entering the game with that on you can press on anything. Quiting that app and entering the game solves the issue.
Thought I mention that here since I had that problem.
