"But I turned it off"
so it won't increase the limit and you have the default limit
Charisma requirements On/Off
On means your max is charisma +10 for population.
Off means you no longer have a max population.
That is what I thought. But I turned it off and verified it is off. I still cannot send a new settler to Sanctuary. The description wording is a little odd which is why I asked. My game still acts like the option is ON. I have to assume some other mod is interfering. Or somewhere is not seeing the updated setting.

Well what I would do is:
1. Save the game.
2. Exit the game.
3. Disable all other mods that affect settlers in a similar fashion.
4. Relaunch the game.
5. Make the attempt to transfer the settler.
If you can find an offending mod then swap the load order. If the conflict still persists then disable the other mod.
Kinggath is really good about his mods not conflicting with others. Speaking of which did you actually go look on the Sim Settlement website in the Framework section?

There is another population limit though which the game uses to decide wether you can send more people over manually (I assume).
The console command setgs iWorkshopSettlerPopulationMax 50 allows me to send more, when that settlement was greyed out before.
Use a bigger number if you really want to go overboard.
getgs iWorkshopSettlerPopulationMax shows the current value, it is 10 by default. So it's gotta be modified in some way to get the actual limit for each location.
There's a mod that makes the setting permanent since changing it with the console won't get saved. (Or you could add the setting to the Fallout4.ini / Fallout4Custom.ini, under [General])

You should give Sim Settlements a try though, I guarrantee it will easily put a lot more hours into the game since you like settlements.
