Report a translation problem
A couple of times I've bought a brahmin from Kelly and they don't show up, presumably they were killed on the way.
I have wondered about Vault 88 though, I would have thought the fact it's sealed off would physically prevent brahmin walking in there, although I know most of these things happen by teleportation anyway.
I bought the third and sent it to Spectacle Island, and it set off on its journey while I was still there. The other two showed no signs of going anywhere.

Did the other brahmin turn up ok at Spectacle Island?
The game sends settlers you move out on their journey at a particular time of day. They might take a day or two to arrive, but it never takes more than one day for them to leave where they've been sent from.
The purchased Brahmin have always started walking away from the trader as soon as the trade was done in every instance I've bought one previously, which is what puzzled me when the first I bought for the vault was still there when I passed again a few game hours later.

And a settlement does not need a Brahmin trough to get them there. They are for keeping the Brahmin from wandering all over your settlements.

If you wanted to handle it with console commands, you could try moving Kelly to the vault and buying the brahmin from her while she's inside. That should rule out any weird pathing issues at least.

So why, when you buy a Brahmin, is the option to send it to Vault 88 offered?

The list includes places where you can't send a Brahmin, and they are greyed out. As the vault is a DLC add-on the devs forgetting to remove it doesn't float. They would have had to add it in the installation of the DLC. The Brahmin trader is in the vanilla game and can't have had the vault option prior to the issue of the DLC.

The only odd part of the whole arrangement is that it doesn't grey out the settlement, but doesn't properly work, either. But there are other cases where they've had similar hiccups in missing just one of multiple settings required to make something work properly.
EDIT: Or it could be a conflict like the vault (incorrectly) being included in the list as a valid choice, but the brahmin AI tries to avoid interior cells. You'd have to see if there's some part of brahmin behavior that's responsible for them not responding to a supposedly valid choice.
