Have to retry several times for the game to work...
I have also very long loading times when entering or exiting places ingame sometimes.
That and having to use a mod for 21:9 support... OMG Bethesda... -_-'
This Exact issue has come up within the last month. The OP had something partially blocking FO4 from launching.
1. Try just verifying the integrity of the game files.
2. Shut down Any unneeded background programs you have running. Recording software, Skype, opened browser, anything that does not need to be running.
3. Try clearing your Download cache;

4. Make sure your antivirus has not updated and is now blocking the game. Add Fallout 4 to the exceptions in it, in Windows Defender and in any other protection software you have.
5. If you have Steam Cloud saving enabled, turn it off. Then the game will save to Documents/My Games Fallout 4. Your issue can sometimes be caused by the game trying to "Sync" when cloud saving is enabled.
EDIT: Since you crash when the intro vids are suppose to start, you could try disabling them. Here's a link to a guide that tells you how. Scroll down to the intro vid part.

But it looks like adding Fallout4.exe to the exception list of my antivirus did the job. I then added f4se_loader.exe to the list afterwards and that's working as well.
