To get a Two-Shot pistol to work, you should use Long Ported Barrel, Muzzle Brake and at least a reflex sight.
Explosive gives you high damage without having to rely on direct hits, and also carries with it no penalty of accuracy.
Two Shot Western Revolvers and .44 might be the exception, as the high base damage increase and the slow ROF can more or less compensate for the two-shot drawbacks.
On a 10mm, two shot is marginally superior but not by much since the second shot is base damage only, but any of an number of things are better suited, VATs Enhanced, Rapid, even things like 'Mutant Slayer' if you know your targets.
That is, if you can control the recoil. Combat Rifles can become unwieldy in Full auto with two-shot, Assault Rifles aren't too bad, and the Handmade Rifle, with Improved Barrel + Muzzle Brake is extremely accurate even in full auto.
The most powerful Two-Shot weapon, however, is the Laser Musket. Six cranks can result in well over 1500 damage in one shot (though it still takes six cells)
Gauss Rifle comes in second, followed by the Railway rifle.
Two shot Plasma rifles are extremely high DPS, but also have incredible recoil problems in automatic mode, so unless you're excellent at burst control, or want it as CQC weapon only (In which it is unrivaled) don't waste your plasma.
Two Shot Laser Rifles, on the other hand, are very underwhelming. You're better off with a Freezing, or ideally, Wounding converted to a scattershot.
I left out Big Boy for obvious reasons.
I would use Explosive if you change your 10mm to an automatic variant, just because the damage output for Explosive automatic or shotgun weapons is so darn huge. Two-Shot is best used on weapons with high base damage, (Gauss Rifle, Railway Rifle, Fat Man, etc.), or other sniping weapons. Explosive and Wounding is best with automatic-firing or shotgun weapons. Instigating is also a good choice for sniper weapons.
