My fave combo was always to take stalwart shell, quick focus, spore mushroom, defenders crest, thorns of agony, grubsong. Doing this you can pretty much just stand in a boss and heal to deal damage whilst losing no health at all :333
You can also use smaller combos of this build like taking quick focus, stalwart, and spore shroom works pretty great
Boss fights, it depends. Stallwart Shell, Quick Slash, grubsong, Quick focus, defender's Crest works for many bosses. Some need a little more finesse.
Probably the best combination when just running arround and exploring, feels fairly broken with how much it trivializes most encounters. Great for bosses too just make sure not to die or you will have to get Fragile Strength again.

*That's it, because most of these charm combos break the game in half. I prefer to keep some challenge. Using any of these charm combos against a boss is basically equivalent to giving up and skipping it.
Baldur Shell would be really good if it actually kept you from getting interrupted while it is being used. Unfortunately, getting hit still interrupts your heal. :(
Similarly, I'd probably use Stalwart Shell more if it also negated knockback from getting hit by enemies, but they can still knock you around.
Quickslash is probably my favorite offensive charm. Being able to swing so quickly makes a huge difference in your damage output, and combined with Mark of Pride it lets you deal with most encounters a lot more quickly and easily.
