I placed 8 AS in Ponyboy and 4 AS in Xyliqil. I believe this setup combined with 'idle' rewarding Ancients will give the most benefits(in terms of HS) in the short-term for a person who just transcended for the first time. But, I didn't use rubies to quik ascent, and so, I'm stuck using the Zombie Auto Clicker to do the hard work manually... manual labor. Currently only reached level 39.
I'm just starting out, but my rule or guideline is to 1) Ascend shortly after I reach the maximum 25 rubies one is able to collect per world, and 2) Transcent when my Hero Souls gains are becoming difficult.
where x is related to your tolerance to grind. i would personally suggest between 3-5. well, obviously not more than 5, that would be immediat transcend.
First transcension I got I think +29, though that was with most progress done before transcension update, so I dunno if that's typical of first transcension or just due to rebalancing factors.
I personally am at my second transcension. My first ever netted me 33 AS and my second was 11AS. I'm currently preparing to transcend again for around 10-12 AS in the next couple of ascensions.

I would say 3 then, 5 seems to be pushing it (not counting getting the next first AS of course).
Three is probably pushing it. I would do it at two.
Btw, after my first transcendence, I am up to level 114... almost 24 hours.
