You either need to do quite a bit of reading up on how trading works in ED, or use a trading tool/website that will tell you where to find good trading routes.
Systems with high demands are usually willing to pay more so if you know that a station buys with a demand of +22% and you know a place that sells at -45% >profit
I'll tell ya what I did to get my Python. (may not be the best way, but it worked for me and quickly)
Buy a Type 6 initially and retrofit it to be light (no weapons) and max cargo space. (I even ditched the shields on mine as well)
But I was able to afford my Python and A rate it in a day or two in my Type 6 a couple weeks ago by just by finding a system in famine and then doing buy & return runs. I would pick missions I knew I could buy multiple commodities from a single station. After you rank up to Friendly/Allied w/ that station, I was easily getting 1-2 million on some missions and completing 2-3 at a time. I think I was cranking out about 10-20 million an hour in my Type 6. (I did upgrade to a Type 7 in the middle of it to double my cargo space... since it was cheap and I didn't need to A rate it other than the FSD)
Use eddb.io to find the closest stations and their commodities to wherever your're doing the missions.
You can find systems in Famine by filtering it in your map.
I didn't buy my Python til I had about 120 million credits, since you really need to get it A rated to make it shine. Also to make sure you have enough for the insurance in case you lose it.
You really need to get your reptuation up with a faction before they'll hand out good paying missions. The good news is that it doesn't take that long. If you stack a bunch of missions from one faction you'll likely get close, if not all the way ranked up.
:edit:
^ Dymhsa covered it. :)
When you're below friendly/allied the payouts suck no matter where you go. But you can rank up your reputation really fast... especially if you take the +++ Reputation reward choice initially over the higher payout.
Usually start popping up once you hit Friendly.
