Rot stink is just a mood penalty until it causes a lung rot infection. The infection can kill, but it takes the infection hediff reaching 100% and the infection still climbs even after you remove someone from the rot stink. As others have said, the main risk is from scaria manhunter events, where lots of dead animals all rot while you're still fighting in the same area.
Tox buildup, meanwhile, is a hediff by itself, and is caused by being in the area of a tox gas cloud or walking over polluted terrain. It stops going up when you leave the causes of toxic buildup, but is itself lethal if it hits 100%.
It's also possible to be immune to tox gas with two detoxifier kidneys, but that doesn't protect you from lung rot. (Although since they're artificial, two detoxifier lungs will make you immune to lung rot.) You can be immune to lung rot with the perfect immunity (to disease) gene, while immunity to tox buildup comes from the dedicated tox immunity gene.
Toxic Buildup is from any toxic source: Tox Gas, Toxic Fallout, Snake Bites, etc.
Rot Stink is from being near any rotting corpses (non-skeletal). You'll see a red miasma seep from dead bodies that release rot stink.
They do not interact with each other in any way.
Yes, and so do things like face masks, but they're only 50% effective by themselves.
I have a shelf full of gas masks (for toxic fallout), it might finally find use for future raids cleanup efforts.
