I am sure if HUNK wasn't a DLC char, he would be the most popular, since he has the best skill set. Gina is relatively popular as starter (and starters seem to be the favoured ones over time), since she has the best healing starter skills.
But AFAIK, the most popular in general is actually Moira. Never understood why, I consider the story mode ending where she finally and actually bits the dust the "good" ending.
1. He's a Russian who's good at sniping and bottle throwing
2. Moira wouldn't had survived without his help
3. As a father he loves his daughter very much
4. He defended his beloved territory to the last breath
I unlocked the Specialist achievement with him.
You might love many others for their one or many (exclusive) skills.
Chris - R.Launcher (to take down Monster Neil) ; Armor of Coat (a true lifesaver)
Hunk - Stealth Cloak (and he comes with 4 open weapon slots + Scavenger skill)
Wesker - Evade Cancel (action breaker)
Cypher - Samurai Sword (for the melee maniacs)
Jill - Can't Catch Me (especially good to go with the Evade Cancel)
the second charactor is Wesker because you need his evade cancel skill, then the third is Chris. You want his rocket launcher.

Edit : rate of fire is crazy and it has Pierce standard cause it's a rifle
