The most random scenario available is the Marko scenario because the rest of the team will be made up of random survivors (who will arrive at random intervals).
The best way to try to create a random customized scenario would probably be to use a list randomizer to pick all your selections/variables/locations.
I like to use only one starting survivor to ensure a random team. I don't like handpicking locations, I like them to be truly random (the order in which certain locations appear is the only factor that makes each play-through slightly different).
I guess another way to make a play-through less predictable/more varied is to select the harshest custom conditions to potentially create some interesting dilemmas..
Note - Choosing an extra long campaign doesn't necessarily increase the difficulty or increase the amount of things to do. The game is balanced for medium length campaigns so anything over 40/50 days you can end up repeatedly fast forwarding every day just to get to the end.
I've never tried one before, but hopefully there are some good ones that don't detail what to expect (in the description) and just leave it for the player to find out.
I agree there should be a button to completely randomize a starting scenario though. Procedural generation adds a lot.
