Can always design a layout on graphing paper. Sometimes it's good to have an idea and build it. Other times you can build to the terrain.
I rarely use PoIs as a main base, mostly just for hordes.
You can really take your preferred design style and create a very defensible base using the building materials provided. The first thing I recommend doing is deciding if you want your "home" base to also be your Blood Moon horde base. If so, then you will be more limited in your design, as you need to insure defensive measures, a strong structure, and sensible zombie pathing. Modularity is definitely possible in any case - you can start small and add on as you acquire resources.
If you want your home base to be separate from your horde base, then you can go all out on the creativity aspect without worrying too much about fending off hordes of zombies. You will, however, still have to provide some defensive measures to cope with the occasional wandering horde and the screamers who are attracted to work station heat signatures.
My personal preference is to set up in a small POI or build a log cabin in early game and reinforce it as needed. I like to build a horde base nearby for the Blood Moons. I eventually move my base deep underground, or even underwater.
Another modular style is the 3x3 pillars, spaced 7 blocks apart. I made a mega base in A17 that sat on a few dozen of these pillars, about 25 blocks off the ground. You can keep the pillars hollow as needed for access up and down to the base. In this pic the 1'st 2 layers are solid steel since I run Parkour.
