With that being said, it comes down to your ammo. I can't remember what ammo types are available for T1 planes, but at higher tiers you can unlock shells for air and ground targets specifically.
Add me if you ever want to squad up and shoot around.
The only exception are very large anti-tank guns (37mm to 75mm) that are mounted on some ground attack aircraft like the German Hs 129. Those do work against aircraft, too, but their rate of fire is too slow to be reliable against them, so they're better off with attacking ground targets.
You're best off using them at the same time for maximum firepower. Once you get machineguns and cannons (20mm and larger), you can think about only using your cannons when you know for sure you're going to hit, because cannons usually have very little ammo available.
You can look this up and change it however you like in the control settings.
I have to assume you are flying one of the P-26's or perhaps some of the P-36's?
So, both machine guns have their purpose-
12.7mm: Extremely hard hitting, probably one of the hardest hitting machine guns in the game. Historically it has been used for just about everything, whether it is anti-personnel, against light vehicles, aircraft, small boats, etc. Personally, I would only fire this at aircraft, as the supply of ammunition given is small, and you get more points for killing other players aircraft.
7.62mm/"30 cal"- These were used on lots of planes in similar calibers, like the .303 British and 7.92mm Mauser, but were ultimately discarded in favor of guns packing a harder punch, namely 12.7mm guns, 13mm guns, or 15-20mm cannons. To me, the biggest value the 7.62mm guns have is killing pilots or setting fires, but the P-26 and P-36A don't have enough 7.62mm's that fire fast enough (For example, the Ishak) for that to be a reliable method of killing.
As for setting different buttons to fire them independantly, I'm not sure that would be worthwhile, as once you run out of 12.7mm the 7.62 doesn't do very much damage at all. As a personal opinion, I would fire both at the same time, and once you have run out of 12.7mm, find a safe place to reload more 12.7mm ammo (Default key "Y", I think).

It depends on both game mode and what your intended targets are- APIT works really good on almost anything, and Stealth works really good.. for well... being stealthy :P
