DS has two DLCs that are not included in DST, those being Shipwrecked and Hamlet. If you get those plus RoG in DS then you will have one content than DST. DS also has Adventure Mode, a story mode that isn't present in DST.
There are people attempting to mod Shipwrecked and Hamlet into DST, but they aren't perfect and far from completed.
Of course, it depends on how much you value a multiplayer experience. If you value that above all else DST is for you.
the ultimate singleplayer experience is a ds game with all dlc worlds merged together.
However, from my recollection, DST is fun enough in single player but a lot of the boss fights and other major events you will still run across at some point or another in dst are a lot harder than they would be in just regular ds. If you think the deerclopse is hard to fight alone in ds good luck soloing it in dst. If i remember correctly things in dst are more or less scaled for multiple people. At least it was that way. Where as ds is the defacto single player dont starve experience. Which as stated before also has things like adventure mode which dst doesnt have. Plus if you have the dlc for dont starve you can do all the little challenges to unlock new characters and play them where as most of the characters in dst have to be purchased.
My personal opinion based off of past experience is to just go with single play ds for, well, single player runs. And do dst for multiplayer. But i would also say it doesnt hurt to try out doing a solo run in dst just to see if it feels right. I just know personally i would go with the route i suggested. But how i feel about any particular game is completely subjective to myself lol
There are mods for DST to balance the bosses for a SP experience but you'll always be playing on the same map. Sure you can get SW and HAM mods for DST, but they wouldn't be officially supported or made by Klei, so bugs and imperfections are obviously things to keep in mind when trying those out.
If you care about challenge or different world/gameplay options for solo play, DS. If you want things to be easier in terms of surviving and don't care much for a change of scenery or gameplay, DST.
