White arrow on buildings?
What is the white arrow that appears when placing buildings? Is this indicating where the building should face a road?

Its the building entrance, but many buildings (most notably houses) never actually make use of the entrance. For production buildings, you do want that arrow pointing toward a road and generally facing toward your warehouse.

I was wondering as well, thank you. Can you actually block the entrance if you're not careful or is it just a matter of "general direction they exit and enter"?

You can block the entrance and villagers will still enter the building. However, walking through buildings decreases the walking speed.
