There are multiple goals you could consider finishing the game:
-Getting the full unlock tree
-Beating P20
-Closing the adamantine Seal
-Beating a queens hand trial
-Completing all steam achievements

But no there is no storyline ending where everyone lives happily ever after.

No rogue lite need an ending.
Ending is self define.
Or else why are dead cell, FTL, into the breach, slay the spire are popular?

If really want a true ending.
Answer is becoming 'queen hand ' and gain audience to the queen

Not quite true, many rogue likes have a story end, but let you start over afterwards. Like enter the gungeons killing the past for each character and ending the gungeon for good. Binding of Isaac's original death ending (and then getting many extra endings). And killing the last boss and escaping the loops for Rogue Legacy (/2), Hades and Loop Hero.

It would be nice to have some kind of conclusion moment for sealing the adamantite seal, even if you have to do p20 to do it.

Well yes, you could consider beating the queens hand that sort of ending. It just takes like 200 hours before you will be ready to even start the queens hand trial and then 50 hours to complete it. So many players will be hoping for an end earlier than that.

Who knows what they might add in the future as dlc or something.
Some new post game adventure with different win conditions would be cool with maybe relating to some of the relics of the past that you can get as house deco.
I found the scarlet orchard biome neato with its fossils so it could be neat with a dlc focused around archeology.

"need" is not applied to rogue lite.
I played the max difficulty in "curse of dead God" and I got the "becoming death champion"ending.
So what?
I click new game and the gameplay still treat me as someone trying to escape from curse, not as a death champion at all.
Because the ending and story is just flavor
The gameplay is core and has nothing to do with your requested ending

Yes. When you die in real life, that's the end.

Well, personally lately I've tried to see just how many embarkation points I can save up realistically (depends very much on the map of the cycle); so I guess this is my kind of "ending". This is my personal best:

Now watch me suddenly die due to something like sudden "blight on the wind" + sealed forest mystery (reduces hostility threshold for events to occur) after loading my caravan with all those juicy delivery lines

The got nice theme going here. I would nice have same base line story fallowing along while you seal those seals. They could expended on lore who is this scorched queen. What seals were for and so on and so on.
Since now most go for endless grind.
